摘 要
中药是我国的宝贵遗产,历史悠久,资源丰富,品种繁多。中药作为国粹,其主要成分的分析以及质量控制已成为世界瞩目的课题。具有分离效率高、速度快、成本低、灵敏度高的、应用范围广的高效液相色谱法在该领域中充当了重要角色。本论文采用反相高效液相色谱法,对一些中草药中的活性成分进行了分离和定量分析,并对其中存在的问题进行了初步探讨。
本论文共分五章:
第一章:对仪器分析特别是色谱法在中草药分析中的应用进行了简单的介绍
第二章:开发了一种利用甲醇-水体系作为流动相的高效液相色谱(HPLC)方法,使马兜铃酸A和马兜铃酸B达到了完全基线分离。该方法简单易行,我们使用这种方法测定了25种广防己和关木通类药物中两者的含量。本文介绍了马兜铃酸的药理作用和结构,研究了色谱条件的优化,并对实验结果进行了分析。
第三章:依据文献内容,采用反相高效液相色谱方法测定了采集于中国和韩国的18种牛膝中蜕皮激素和齐墩果酸含量。该方法分别采用乙腈-水混合流动相以及甲醇-水混合流动相快速测定了牛膝中蜕皮激素和齐墩果酸的含量,两种方法的色谱峰面积和保留时间的重现性很好,灵敏度很高,是测定蜕皮激素和齐墩果酸的简便而又准确的方法。我们通过对实验结果的比较,得出了牛膝种属和产地对蜕皮激素和齐墩果酸含量的不同影响,相对于产地而言,种属是主要因素。
第四章:使用反相液相色谱法测定了10种不同地点采集的翼首草中齐墩果酸和熊果酸的含量。该方法以甲醇-水-乙酸体系作为流动相,操作简便,成本低,重现性好,这两种物质的工作曲线相关系数分别是0.9998和0.9994。本文主要进行了实验条件的优化,比较了通过不同提取方法得到样品的测定结果,并对实验结果进行讨论,得到了不同产地和不同部位的翼首草中该成分含量的差异。也对翼首草药物的用药原理进行了简要的概括。
第五章:在高效液相色谱-电化学检测器联用测定酪氨酸和多巴胺的过程中,对碳纳米管材料涂布后的玻碳电极和未涂布的电极在灵敏度、重复性等方面进行了比较。

Abstract
Traditional Chinese medicines with long history and various kinds are valuable
treasures of Chinese people which have attracted interest from most countries
in the word because of their special effects.In recent years,the effective methods
for quality control of the herbal medicines has become the focus of analysis
of Chinese herbs.High performance liquid chromatography (HPLC) characterized
with high separate efficiency,low cost,rapidity,far-range using and high sensitivity
plays an important role for analysis of Chinese herbs. The thesis, deals with
the separations and quantity determinations of some effective components in
several kinds of Chinese herbal medicines.
This thesis is composed of five parts.
Part 1:This chapter is a comprehensive review on the applications of instrumental
analysis especially chromatography on analysis of Chinese herbal medicines.
Part 2:A reverse phase high-performance liquid chromatographic method has been
developed for the determination of ristolochic acid A and aristolochic acid
B in 25 samples of fangji and 。Baseline seperation was achieved using ODS C18
column and mobile phase composed of methanol and water. Two kinds of aristolochic
acid in 25 samples of fangji or 关木通 were successfully determined using this
method Their strctures ,medical functions and the optimazition of experimental
conditions are discussed in this part.
Part 3:Determinations for oleanolic acid and ecdysterone in 18 samples of Achyranthes
bidentata blume collected from Korea and China by HPLC methods using mobile
phases composed of acetonnitrile and water or methanol and water.The two methods
were proved to be complementary methods with rapidity、high sensitivity and good
repeatability. This Part introduces the experimental conditions and deals with
the result.The categories of Achyranthes bidentata blume have more effections
on the qulities of oleanolic acid and ecdysterone than their producing areas.
Part 4:This part is the emphases of this thesis. Oleanolic acid and ursolic
acid in 10 samples of Pterocephalus hookeri from different areas and altitudes
were determined by a HPLC method using mobile phases composed of methanol and
water and acetic acid. This method is precise simple and cost-effective.The
correlation coefficients of calibration curves of oleanolic acid and ursolic
acid were 0.9998 and 0.9994. This part discusses the optimazition of experimental
conditions and compares two kinds of extractions. The results shows that contents
of oleanolic acid and ursolic acid varied significantly from locality to locality,from
low altitude to high altitude.Different positions of Pterocephalus hookeri have
different contents of oleanolic acid and ursolic
