【摘要】 目的 旨在探讨在体外胰岛素能否促进人肝癌细胞增殖,而奥曲肽对其诱导的增殖活动有无抑制作用。方法 将奥曲肽与胰岛素直接作用于体外培养的人原发性肝癌细胞株(BEL-7402),运用MTT、直接细胞计数及流式细胞仪测定等方法观察奥曲肽与胰岛素对(BEL-7402)肝癌细胞的活细胞数及其比值、细胞总数、细胞周期及增殖指数(PI)的影响。结果 胰岛素(0~5μg/ml)使细胞株的细胞总数、活细胞数及其比值增加;呈明显的量效关系,而奥曲肽(0~2μg/ml)则使(BEL-7402)细胞基础的及胰岛素刺激的细胞总数、活细胞数及其比值降,亦呈明显的量效关系。另外,5μg/ml的胰岛素增加细胞的PI值而1μg/ml的奥曲肽降低其PI值(P<0.05),并对BEL-7402细胞株产生S期阻滞作用。结论 在体外,胰岛素可诱导人肝癌细胞增殖,奥曲肽不仅直接抑制肝癌细胞的增殖,对胰岛素诱导的肝癌细胞增殖也有抑制作用。

A study of effect of Octreotide and insulin on human hepatoma cell in vitro

【Abstract】 Objective To investigate in vitro whether the proliferative action of human hepatoma cell can be induced by insulin, which can be inhibited by Octreotide . Methods A human hepatoma cell line BEL-7402 was used in this study . Using MTT method , cell count and flow cytometry (FCM), the action of insulin (0~5μg/ml)and/or Octreotide (0~2μg/ml)on the number of viable cell as well as its ratio, the total number of cell, cell cycle and proliferative index (PI) in phase –synchronized cells were assayed. Results The cell number, the viable cell number and its ratio in insulin (0~5μg/ml)-treated BEL-7402 cells increased while decreased in basic and insulin-induced ones treated by Octreotide (0~2μg/ml), both in dose-dependent manner. 1μg/ml of Octreotide resulted in a reduced PI which was increased in cells treated by 5μg/ml of insulin. Moreover , a S cell cycle block was detectable in phase-synchronised cell subjected to treatment of 1μg/ml of Octreotide. Conclusions In vitro, the growth proliferation of human heptoma cell can be induced by insulin, while Octreotide can exert an inhibitory effect on basic and insulin-induced growth in human hepatoma cell.
